چکیده

Introduction MicroRNAs may be implicated in the acquisition of drug resistance chronic myeloid leukemia as they regulate expression not only BCR-ABL1 but also genes associated with activation transfer proteins or essential signaling pathways. Methods To understand impact specifically expressed miRNAs and their target genes, we collected peripheral blood mononuclear cells (PBMC) from patients diagnosed (CML) healthy donors to determine whole miRNA by small RNA sequencing screened out 31 differentially microRNAs (DE-miRNAs) high expression. With utilization set enrichment analysis tools, present here a comprehensive relevance DE-miRNAs disease biological function. Furthermore, literature-based miRNA-target gene database was used analyze overall define responses. We further integrated DE-miRNA identify CML targeted signature singscore (CMTGSS) gene-set (GSEA) correlation between CMTGSS Hallmark gene-sets PBMC samples clinical patients. Finally, association stratification multiple cell lineage sets validated using GSEA. Results Although individual have been reported varying degrees on CML, overall, our results show that abnormally upregulated are apoptosis aberrantly downregulated cycle. The shows defined prognosis CMTGSS-based presented tendency for affect differentiation microenvironment. Conclusion Collectively, this study comprehensively analyzed functions differential genes. specific differentiated immune suggests magnitude potential targets setting. It helps us make links different obtained multi-faceted studies provide more research directions.

Expression Profiling of Microarray Gene Signatures in Acute and Chronic Myeloid Leukaemia in Human Bone Marrow

Background Classification of cancer subtypes by means of microarray signatures is becoming increasingly difficult to ignore as a potential to transform pathological diagnosis nonetheless, measurement of Indicator genes in routine practice appears to be arduous. In a preceding published study, we utilized real-time PCR measurement of Indicator genes in acute lymphoid leukaemia (ALL) and acute m...

Expression profiling of peripheral-blood mononuclear cells from patients with chronic hepatitis C undergoing interferon therapy.

BACKGROUND Interferon (IFN) is now the standard treatment for chronic hepatitis C (CH-C); however, treatment efficacy is unpredictable before IFN therapy is started. METHODS We investigated the gene-expression profiles of peripheral-blood mononuclear cells (PBMCs) from patients with CH-C showing different responses to IFN. Gene-expression profiles of PBMCs were analyzed in 21 patients with CH...

Toll Like Receptor 2 and 4 Expression in Peripheral Blood Mononuclear Cells of Multiple Sclerosis Patients

Background: Multiple sclerosis (MS) is a T cell mediated autoimmune disease with unknown etiology. Appropriate MS therapeutic strategies need thorough understanding of both disease etiology and pathogenesis mechanisms. Ligation of TLR-2 and TLR-4 stimulates the production of several cytokines leading to CNS autoimmunity and neurodegenerative diseases. Objective: To find a relationship between M...

Conditioned Medium from Cultured Colorectal Cancer Cells Affects Peripheral Blood Mononuclear Cells Inflammatory Phenotype in Vitro

Background: Colorectal cancer (CRC) is the third most common cancer worldwide. Studies have indicated that immune cells and soluble factors play a key role in maintaining the balance between tumor-promoting inflammation and anti-tumor immunity. It has been shown that secreted cytokines from CRC cell lines could affect peripheral blood mononuclear cells (PBMCs), monocytes, and macrophages phenot...
